Practical Application

Does one big session build fitness, or the weeks of them? The weeks. Each session sends a brief molecular signal; mitochondria and capillaries grow from repeated sessions with recovery between, and both volume and intensity count.
What it means for training: weekly time and frequency carry the dose (SYNTH-51); intensity buys more mitochondrial gain per hour (REF-465, SYNTH-26).
What it does not mean: that one zone is the only mitochondrial stimulus (Zone 2 is not shown to be optimal, REF-1092), or that a bigger acute signal means a bigger gain (signal and adaptation dissociate, as train-low shows, SYNTH-83).

Mechanism

A session's metabolic perturbation activates signalling that drives transcription and translation, and repeated bouts remodel the muscle (REF-457); repeated energy stress promotes mitochondrial biogenesis through hormetic feedback loops (Perry & Hawley 2018); calcium and AMPK signalling converge on PGC-1α (REF-463, full text).
Over seven interval sessions, PGC-1α mRNA rose more than tenfold 4 h after the first and was back to control within 24 h, a saw-tooth that preceded the rise in mitochondrial proteins (Perry et al. 2010, one biopsy study).
Mitochondrial content rose 23% with endurance and 27% with interval or sprint training, more with more sessions a week; per hour, intervals and sprints gave about 1.7 and 3.9 times the endurance gain (REF-465, 5,973 participants).
High-dose vitamins C and E blocked the rise in mitochondrial markers while VO2max rose alike (REF-908, 54 young adults).

Common misreading

"Zone 2 is the only way to build mitochondria"; "one epic ride makes up for a missed week".
